چکیده
This study was conducted to investigate the metabolic defensive mechanism in the larvae of African palm weevil (Rynchophorus phoenicis) administered with dichlorvos (2,2-dichlorovinyl dimethylphosphate) solution. Bioassay experiment with dichlorvos was conducted on the larva and glutathione-utilizing enzyme activities were determined in the major organs: fat body, gut, and head of R. phoenicis larva 48 h after treatment with 0–0.060 μg g−1 body weight dichlorvos solution. Glutathione transferase was purified from the gut of larvae by ion-exchange chromatography on diethylaminoethyl-Sephadex A50 and affinity chromatography on glutathione-Sepharose 4B columns. The purified enzyme was homogenous as revealed by sodium dodecylsulfate polyacrylamide gel electrophoresis. Initial velocity studies were carried out on the purified enzyme using standard procedures. Bioassay experiment indicated alterations of glutathione peroxidase, glutathione reductase, and glutathione transferase activities in the major organs of larva caused by dichlorvos. Glutathione transferase activity in the gut of larva was three times higher than that of glutathione peroxidase and glutathione reductase activities, an indication of possible detoxification role of glutathione *Corresponding author: Olufemi Samuel Bamidele, Enzymology Research Unit, Department of Biochemistry, The Federal University of Technology, P. Chemical Evaluation of African Palm Weevil, Rhychophorus phoenicis, Larvae as a Food Source
The chemical properties of the African palm weevil, Rhychophorus phoenicis (F.) (Coleoptera: Curculionidae), larvae were evaluated using standard methodology. The chloroform-methanol extract yielded 37.12% on a dry basis. The oil was liquid at room temperature with a flash point of 36.0 °C. Analysis of the physical constants indicated values of 192.25 Wijs and 427.70 mg KOH/g as iodine and sapo...

This study aimed to explore the larvicidal and growth-inhibiting activities, and underlying detoxification mechanism of red palm weevil against phenylpropanoids, an important class of plant secondary metabolites. Toxicity of α-asarone, eugenol, isoeugenol, methyl eugenol, methyl isoeugenol, coumarin, coumarin 6, coniferyl aldehyde, diniconazole, ethyl cinnamate, and rosmarinic acid was evaluate...
متن کاملAntioxidative Response in a Silkworm, Bombyx mori larvae to Dichlorvos insecticide
Introduction: Antioxidant defense components protect insects by scavenging reactive oxygen species, leading to oxidative stress. We therefore investigated the effect of an organophosphate insecticide, dichlorvos, on superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT), glutathione S-transferase (GST) and glutathione reductase (GR) as antioxidative biomarkers in silk worm, Bo...
متن کاملLipase Activity in the Larval Midgut of Rhynchophorus palmarum: Biochemical Characterization and the Effects of Reducing Agents
Lipases have key roles in insect lipid acquisition, storage, and mobilization and are also fundamental to many physiological processes in insects. Lipids are an important component of insect diets, where they are hydrolyzed in the midgut lumen, absorbed, and used for the synthesis of complex lipids. The South American palm weevil Rhynchophorus palmarum is one of the most important pests on comm...
متن کاملSpinosad Induces Antioxidative Response and Ultrastructure Changes in Males of Red Palm Weevil Rhynchophorus ferrugineus (Coleoptera: Dryophthoridae)
The red palm weevil, Rhynchophorus ferrugineus, is of great concern worldwide, especially in the Middle East, where dates are a strategic crop. Despite their ecological hazard, insecticides remain the most effective means of control. A bioinsecticide of bacterial origin, spinosad is effective against several pests, and its efficacy against male R. ferrugineus was assessed in the present study. ...
